疏青柿果中柿单宁的最佳提取工艺

Extraction technology of persimmon tannin from hand-picked green persimmon

  • 摘要: 采用超声波辅助法与酶解法相结合,以80%酸性甲醇为提取液提取疏青柿果中的柿单宁,用Folin-Ciocaileu法测定总酚含量,用香草醛−硫酸法测定柿单宁含量,并通过单因素实验和正交实验探究不同条件对柿单宁含量和产品得率的影响。结果显示,柿单宁提取的最佳工艺条件为纤维素酶的添加量为0.5 g,提取料液比为1∶15,盐酸浓度为1%,提取时间为90 min,提取温度为80 ℃。经验证实验测得此条件下提取物总酚含量、提取物缩合单宁含量和产品得率分别为29.13 mg/g、22.33 mg/g、7.08%。采用正交实验法优化疏青柿果中柿单宁的提取工艺稳定、高效、可行。

     

    Abstract: In the study, 80% acidic methanol was used to extract the tannin of hand-picked green persimmon by the ultrasonic-assisted method combined with enzymatic hydrolysis. The total phenolic content was determined by Folin-Ciocaileu method, and the tannin content was determined by the vanillin-sulfuric acid method. The effects of different conditions on the tannin content and yield of persimmon fruit were investigated by single factor experiment and an orthogonal experiment. The results show that the optimal extraction conditions are as follows: the addition of cellulase is 0.5 g, the ratio of material to liquid is 1∶15, the concentration of hydrochloric acid is 1%, the extraction time is 90 min, and the extraction temperature is 80 ℃. The total phenolic content, condensed tannin content and yield of the extract are 29.13 mg/g, 22.33 mg/g, and 7.08%, respectively. The orthogonal experiment was used to optimize the extraction process of tannin from hand-picked green persimmon, demonstrating stability, efficiency, and feasibility.
